ARA-290 is an 11-amino acid synthetic peptide derived from the helix B surface of erythropoietin, studied for neuroprotective and anti-inflammatory properties without EPO's hematopoietic effects.
ARA-290 (also known as cibinetide) is a synthetic peptide corresponding to residues on the helix B surface of erythropoietin (EPO). While EPO is best known for its role in red blood cell production, research has identified a separate receptor system — the innate repair receptor (IRR) — through which EPO exerts tissue-protective effects independent of hematopoiesis. ARA-290 was developed to selectively engage this IRR pathway without triggering erythropoiesis.
The innate repair receptor is believed to be a heterodimer of the EPO receptor and the beta common receptor (βcR). ARA-290 binds selectively to the IRR without activating the classical homodimeric EPO receptor responsible for red blood cell production. This selectivity makes it a useful research tool for studying tissue-protective EPO signaling in isolation from hematopoietic effects, which has historically complicated EPO research.
Beyond neuroprotection, ARA-290 has demonstrated anti-inflammatory activity in preclinical studies. It has been shown to modulate macrophage polarization toward an anti-inflammatory phenotype and reduce pro-inflammatory cytokine release in LPS-stimulated cell cultures. These properties have generated research interest in ARA-290 as a tool for studying inflammatory disease mechanisms, particularly in conditions with a neuropathic component. All NuvoTide ARA-290 is supplied for in-vitro research use only.
